Research Technician III - Virology Lab The Silva Lab at the University of Pittsburgh is seeking a highly motivated, reliable, and detail-oriented Research Technician to be an integral part of a dynamic research team, conducting independent and collaborative research in a virology-focused laboratory with a diverse and cooperative atmosphere. We study various facets of chikungunya virus, a mosquito-borne virus that causes a severe febrile illness characterized by acute and chronic musculoskeletal manifestations. The position will include performing supervised and independent research at BSL2 and A-BSL3 conditions consisting of 1) tissue culture and virological assays; 2) molecular biology-based assays to uncover mechanisms of viral replication; and 3) pathogenesis studies using murine models of disease.The incumbent will provide research support to other members of the team and will manage an independent research project, given sufficient training, familiarity, and proficiency. Primary scientific tasks will include conducting complex lab experiments, helping to develop research methods and procedures, maintaining detailed experimental notes, and organizing data and laboratory samples. Responsibilities will include virus stock generation and characterization, cell culture, DNA/RNA extraction, bacterial cloning, RT-qPCR and standard PCR, protein analysis (SDS-PAGE), and basic virological assays. Prior working experience in virological assays is preferred but not required. In addition, the technician will be trained to work in our A-BSL3. Applicants should expect to be comfortable wearing a powered air purifying respirator (PAPR) and other personal protective equipment. The technician will assist with animal colony breeding, maintenance, and animal experiments, which will include infections, tissue harvests, and histological sample preparation in the A-BSL3. Prior working experience with animals is required. Previous work in a BSL3 would be considered highly advantageous, but on-the job training will be provided. Strict adherence to BSL2/BSL3 procedures is essential. The research technician will work closely with the PI and members of the laboratory to ensure that experiments are conducted according to lab protocols.Other responsibilities for this position include performing common lab duties, such as general lab maintenance, ordering and maintaining lab inventory of supplies and reagents, making common stock solutions, maintaining equipment in working order, and routine duties and maintenance of the A-BSL3 facility. The ideal candidate should have knowledge of basic laboratory skills and must be able to work independently on tasks after training. This position calls for efficiency, creativity, multitasking, and the desire and ability to effectively conduct independent studies in a team setting.
